Irritation testing is often used to evaluate the potential irritation of biological materials, medical devices, or products exposed to the eyes, skin, or mucous. It is usually using a model appropriate for the route and duration of exposure or contact. Human oral mucosa has many chemical and physical barriers to transmucosal absorption to avoid toxicity. In the development process of oral care products, it is particularly important to evaluate the potential of oral irritation and identify substances that may cause adverse oral reactions. Oral irritation testing greatly reduces the potential risks of oral care products and provides valuable safety data for products that directly contact the oral epithelium, including toothpaste, mouthwash, and medical devices.

Based on normal human-derived buccal cells, tissue-engineered 3D culture systems for oral mucosa have been developed. These cells are cultured to form a highly differentiated human oral tissue 3D model. These models have been widely used to study the response of human oral mucosa to external stimuli in vitro, and become an efficient alternative to expensive clinical or whole animal testing. In this test method, the finished product or component being tested is directly applied to the surface of the tissue, providing a good model of "real life" exposure.

The 3D model of highly differentiated human oral tissue is composed of highly organized basal cells, Stratum Filamentosum and Stratum Distendum. From the morphological comparison, the oral epithelial model has a uniform thickness and is very similar to native buccal tissue. The tissue model grows on a special platform at the air-liquid interface, which can accurately simulate "real-life" oral exposure, thereby helping researchers perform test projects.

Creative BioMart has accumulated a lot of oral irritation test experience, and can perform various toxicology test services on our powerful platform to help personal care product manufacturers comprehensively evaluate their products. We evaluate the potential irritation of the product based on the "ET50" value, that is, the time (in minutes) it takes for the product to reduce the cell viability of the tissue model to 50%. Cell viability is measured by enzymatically converting the vital dye MTT into a blue formazan salt, and then the ET50 value is used to specify the potential irritation level of a series of products or ingredients.

In addition, we also provide pro-inflammatory cytokines to induce inflammation in the test system to investigate the protective or anti-inflammatory effects of test items.
