This kind of non-regulatory test, also called lung irritation test, can provide information on the damage of the test substance to the lining of the human airway within a certain concentration range. Inhalation toxicity can be specifically defined as the degree to which the test item causes damage (toxicity) to human airway after being exposed to a single substance or mixture for up to 4 hours. It is an important part of product safety assessment and an important part of the occupational safety of manufacturing workers. Various volatile chemical components may cause inhalation toxicity, including cosmetic fragrances, fuels, adhesives, and nanoparticles, etc.

Our Services

We use the 3D human airway tissue model (EpiAirway) to simulate the biochemical and physiological characteristics of the human respiratory epithelial tissue. These cells have been cultured to form a multi-layer highly differentiated model, which consists of a pseudostratified epithelial structure complete with tight junctions as well as microvilli, cilia, and mucin secretion at the apical surface, and very similar to the tracheal/bronchial epithelium in normal humans. EpiAirway grows on the platform of the air-liquid interface, and can accurately simulate the manner of "real life" airway exposure in the test. In some cases, caps are used to prevent excessive evaporation of volatile substances.

We measure cell viability by enzymatically converting the vital dye MTT into blue formazan salt, and then perform quantitative measurement after extracting the salt from the airway tissues. Toxicity is determined by calculating the IC75 value, which is the concentration of the test item required to reduce the viability of the airway model to 75%. If the IC75 value is below the threshold, the test item is used to classify it as toxic.
