In Vitro Skin Penetration Test

The skin acts as a barrier and is responsible for protecting humans from the substances contacted in the environment. The penetration of a substance depends on many factors, such as the formulation presented to the skin; the physicochemical properties of the test substance, including lipophilicity, molecular weight, charge, and concentration; and the area and time of exposure.

Quantitative data on the bioavailability of compounds applied to the skin is now receiving increasing attention, not only in the pharmaceutical industry but also in the cosmetic industry. Skin penetration testing, also known as percutaneous penetration, measures the absorption or penetration of a substance through the skin barrier and into the skin (OECD, 2004a). This test can determine how much of the chemical penetrates the skin, thereby further determining whether it has the potential to be absorbed into the systemic circulation. In addition to efficacy studies, the rate and extent of dermal and transdermal absorption are also closely related to the safety evaluation of various xenobiotics, such as sunscreens and pesticides.

It should be noted that EU Regulation (EC) 1223/2009 no longer allows skin absorption testing of new cosmetic ingredients in live animals, so there is a need for effective alternatives in cosmetic safety testing. Currently, the use of in vitro models to test cosmetic ingredients has become the norm.
