Effect of growth factors on skin

Growth factor (GF) is a cytokine that regulates the growth and development of different cells by binding with specific cell membrane receptors in vivo. It can prevent skin aging by stimulating collagen and elastin, and trigger the skin's innate healing ability. The most important GF involved in skin proliferation and remodeling are as follows:

1) Epidermal Growth Factor: Epidermal growth factor can promote cell proliferation, promote the synthesis of hyaluronic acid and elastic fibers, achieve skin rejuvenation, increase the transport of nutrients and sugar metabolism, thereby reducing and eliminating wrinkles. It is worth mentioning that it can also induce reverse differentiation of stem cells and repair wounds.

Growth factors can be used in hydrogel, lotion, essence or injection form. Growth factors have been used many times in the medical application of wound healing and angiogenesis. These products are in injection or topical forms using hydrogel or stent systems. Because of its simple composition and high concentration ability, water-based essence dominates the cosmetics anti-aging market of EGF products for external use. Clinical studies have shown that the application of growth factor has certain anti-aging effects, such as repairing, eliminating wrinkles, anti-aging, lightening stains and moisturizing. First, GF can be absorbed through sweat glands and hair follicles. Secondly, as the aging skin is drier and more fragile, even if it is intact, it is more vulnerable to the penetration of irritants and larger molecules. Finally, there is paracrine communication between keratinocytes and fibroblasts, and keratinocytes can induce fibroblasts to produce GF. The penetration of a small amount of locally applied GFs in the epidermis may promote GFs from platelets (TGF-α and TGF-β). It secretes and promotes the activation and proliferation of dermal fibroblasts, thus remodeling and regeneration of the extracellular matrix.

Other characteristics of growth factors

  • The integrity of growth factors may be affected by pH and temperature, etc. Pay attention to storage conditions during application. The optimal pH ranges from 6.0 to 8.0, and the isoelectric point is about 4.6.
  • One of the important factors affecting the stability of the growth factor formula is the interaction between solvent, excipient and ionizable group of the protein side chain. EGF is hydrophilic in nature and requires aqueous solvents. A final product concentration of 1 μ G/30g is considered appropriate and stable.
  • Reducing sugar has the largest negative impact on the stability of EGF. PEG 6000 and polysorbate 20 and 80 have been proven to increase oxidation.
